A systems-based approach to ensure berry crops health status: from the breeder to the field
Introduction
The term berries refers to strawberry, blueberry, blackberry, raspberry and their hybrids e.g. loganberry, boysenberry, cranberry, currants, gooseberry, elderberry, honeysuckle and huckleberry (members of the genera Fragaria, Rubus, Ribes, Vaccinium, Sambucus and Lonicera). Berries have been harvested from the wild since the onset of human evolution. It was not until the development of modern strawberry (F. x ananassa) in the eighteenth century that berry breeding came into the horticultural forefront (Darrow, 1966). Because of their nutritional value, berries have become a leading food in many countries (Nile & Park, 2014), accounting for approximately $10 billion in fresh market global production (FAOSTAT, 2023), with tens of new cultivars developed and released every year to address the consumer need for flavor, appearance, size, shelf life and adaptability to new environments (Edger et al., 2022; Foster et al., 2019; Senger et al., 2022).

Abstract
Photorhabdus and Xenorhabdus species have mutualistic associations with nematodes and an entomopathogenic stage1,2 in their lifecycles. In both stages numerous specialized metabolites are produced that have roles in symbiosis and virulence3,4. Although regulators have been implicated in the regulation of these specialized metabolites3,4, how small regulatory RNAs (sRNAs) are involved in this process is not clear. We show here that the Hfq-dependent sRNA, ArcZ, is required for specialized metabolite production in Photorhabdus and Xenorhabdus. We discovered that ArcZ directly base-pairs with the mRNA encoding HexA, which represses the expression of specialized metabolite gene clusters. In addition to specialized metabolite genes, we show that the ArcZ regulon affects ~15% of all transcripts in Photorhabdus and Xenorhabdus. Thus, the ArcZ sRNA is crucial for specialized metabolite production in Photorhabdus and Xenorhabdus species and might well become a useful tool for metabolic engineering and identification of commercially relevant natural products.
